  • 종설명(Description)

Echidna nebulosa A single specimen (522.0 mm in total length) of the family Muraenidae, collected from the waters off south-western Jeju-do Island in November 2021, was identified as Echidna nebulosa on the basis of morphological and molecular methods (DNA barcoding). This species is characterized by several morphological traits as follows: 10 rounded molar, median intermaxillary, and vomerine teeth; 4 predorsal vertebrae, 50 preanal vertebrae and 123 total vertebrae; and a pale background color with two rows of 17-20 blackish brown starry blotches along the head and body. As this is the first record of E. nebulosa in Korea, we propose the new Korean names, 'Dung-Geun-Ni-Gom-Chi-Sog' for the genus Echidna, and 'Beol-Kkok-Gom-Chi' for E. nebulosa. 강충배,이유진,김진구,한송헌 한국수산과학회 상세보기
Differences and Variations among Anguilla japonica, Muraenesox cinereus and Conger myriaster from the Yellow Sea Genomic DNAs were extracted from the muscle of twenty-one specimens of three eel species collected in Anguilla japonica (AJ), Muraenesox cinereus (MC) and Conger myriaster (CM) from the Yellow Sea, respectively. In the present study, 7 oligonucleotides primers generated 191 specific loci in the AJ species, 226 in the (MC) species and 181 in the CM species, respectively. The primer BION-02 generated the most loci (a total of 83), with an average of 11.86 in the AJ species. The specific loci generated by oligonucleotides primers exhibited inter-individual-specific characteristics, thus revealing DNA polymorphisms. With regard to average bandsharing value (BS) results, individuals from Conger myriaster species (0.808) exhibited higher bandsharing values than did individuals from Muraenesox cinereus species (0.729) (P<0.05). The longest genetic distance (0.430) displaying significant molecular difference was also between individual no. 01 within Anguilla japonica eel species and individual no. 04 within Anguilla japonica species. In this study, the dendrogram resulted from reliable seven oligonucleotides primers, indicating three genetic clusters composed of group I (ANGUILLA 01~ANGUILLA 07), group II (MURAENESOX 08~MURAENESOX 14) and group III (CONGER 15~CONGER 21). The existence of species differentiation and DNA polymorphisms among three eel species were detected by PCR analysis. As mentioned above, a dendrogram revealed close relationships between individual identities within three eel species. High levels of a significant genetic distance among three eel species showed this PCR approach is one of the most suitable tools for individuals and/or species biological DNA studies. Differences in organochlorine accumulation accompanying life history in the catadromous eel Anguilla japonica and the marine eel Conger myriaster

In order to examine the ecological risk for organic pollutants in diadromous fish migrating between sea and freshwater, organochlorine compounds (OCs) were determined in the catadromous eel Anguilla japonica having marine, estuarine and freshwater residence life histories. The eels were collected in Japanese coastal areas. We also compared the OCs accumulation with the marine eel Conger myriaster, which has a similar life history as A. japonica in the marine environment. The ontogenic changes in the otolith strontium (Sr) and calcium (Ca) concentrations were examined along the life history transect to discriminate the migration type. There were generally three different patterns, which were categorized as 'marine residence' (spent most of their life in the sea and did not enter freshwater), 'estuarine residence' (inhabited estuaries or switched between different habitats), and 'freshwater residence' (entered and remained in freshwater river habitats after arrival in the estuary) according to the otolith Sr:Ca ratio. There were generally no correlations between OCs such as Dichlorodiphenyltrichloroethanes (DDTs), Hexachlorobenzene (HCB), Hexachlorocyclohexanes (HCHs) and Chlordanes (CHLs) accumulation and each biological characteristic such as TL, BW and age in A. japonica. A positive correlation between the lipid content and concentrations of OCs were found. Additionally, the concentrations of HCB, HCHs, CHLs and DDTs in A. japonica were significantly higher than those of C. myriaster, associating with the higher lipid contents (14% on average) in the former than the latter eels (9% on average). A negative linear relationship was found between the otolith Sr:Ca ratios and concentration of each OCs in A. japonica. The ecological risk of OCs increase as the freshwater residence period in the eel becomes longer. It is clear that migratory histories and lipid contents directly affected OCs accumulation in the anguillid eels.

Cloning and characterization of vitellogenin cDNA from the common Japanese conger (Conger myriaster) and vitellogenin gene expression during ovarian development

        Abstract

The major yolk protein precursor, vitellogenin (VTG) was detected in plasma from vitellogenic females and estradiol-17β (E2)-treated immature females, but not in males and immature females by Western blotting in common Japanese conger Conger myriaster. Its molecular mass was approximately 180 kDa under denaturing and reducing conditions. The common Japanese conger VTG cDNA was cloned from the liver of vitellogenic female. It contains 5110 nucleotides including an open reading frame that encodes 1663 amino acids. The deduced amino acid sequence of the common Japanese conger VTG shares 80% identity with that of eel Anguilla japonica VTG-1, and 45?55%, 32?34% and 27?29% identity with the deduced amino acid sequences of other fish, amphibian and avian VTG with polyserin domain, respectively. In female common Japanese conger, VTG gene was highly expressed in the liver of this species similar with other oviparous vertebrates. The expression levels of VTG gene in the liver increased from the oil droplet stage to the tertiary yolk globule stage and were maintained until the migratory nucleus stage.
